Matthaei nació en [[Kensington]] nuna familia d'ascendencia alemana, suiza y francesa. Yera fía del comerciante comisionista Carl Hermann Ernest Matthaei y de la música Louise Henriette Elizabeth Sueur, y tenía un hermanu y trés hermanes más pequeñes, incluyendo a Louise Howrard.<ref name="odnb">{{Obra citada|title = Howard, Louise Ernestine|last = Oldfield|first = Sybil|work = [[Oxford Dictionary of National Biography]]|publisher = [[Oxford University Press]]|url = http://www.oxforddnb.com/view/article/37576|accessdate = 1 de xineru de 2013}}</ref> Matthaei asistió a la escuela pa moces North London Collegiate School y a Newnham College, Cambridge, y depués trabayó como asistente de [[Frederick Frost Blackman|Frederick Blackman]], produciendo importantes trabayos sobre [[respiración celular]].
