El so verdaderu nome yera Lewis Michael Arquette, y nació en Chicago, Illinois. Los sos padres yeren Mildred Nesbitt y l'actor Cliff Arquette.[3] Yera descendiente del esplorador Meriwether Lewis,[4] y foi parte de la familia d'intérpretes Arquette. Amás del so padre, Cliff Arquette, otros miembros de la familia son los sos fíos Patricia, Alexis, Rosanna, David y Richmond Arquette. Foi'l suegru de l'actriz Courteney Cox y del actor Thomas Jane, y antiguu suegru del tamién actor Nicolas Cage. Arquette actuó con frecuencia xunto a los sos fíos.
Mientres vivía en Chicago, Arquette dirixó dellos años el teatru The Second City. En 1970 la familia camudar a una comuña en Front Royal, Virxinia.
